ABSTRACT

Objective To investigate the effect of long intergenic non-coding RNA COX2 (lincRNA-COX2) on apoptosis and polarization of Listeria monocytogenes (Lm)-infected RAW264.7 cells. Methods RAW264.7 cells were cultured and divided into control group (uninfected cells), Lm infection group, negative control of small interfering RNA (si-NC) group, si-NC and Lm infection group, small interfering RNA of lincRNA-COX2 (si-lincRNA-COX2) group, si-lincRNA-COX2 and Lm infection group. RAW264.7 cells were infected with MOI=10 Lm for 6 hours, and then the inhibition efficiency of siRNA transfection was detected by fluorescence microscope and quantitative real-time PCR (qRT-PCR). The expression levels of cleaved-caspase-3(c-caspase-3), caspase-3, B-cell lymphoma-2 (Bcl2), Bcl2 associated X protein (BAX), arginase 1 (Arg1), inducible nitric oxide synthase (iNOS) were detected by Western blot analysis. Results c-caspase-3/caspase-3, BAX/Bcl2 and iNOS were significantly up-regulated, while the level of Arg1 was down-regulated in Lm-infected RAW264.7 cells compared with control group. LincRNA-COX2 knockdown inhibited the increase of protein levels for BAX/Bcl2, c-caspase-3/caspase-3 and iNOS in Lm-infected RAW264.7 cells, while the level of Arg1 in Lm-infected RAW264.7 cells was up-regulated. Conclusion Knockdown of lincRNA-COX2 can inhibit cell apoptosis and suppress the macrophage polarization into M1 type in Lm-infected RAW264.7 cells.

ABSTRACT

A bactéria Listeria monocytogenes é o patógeno causador da listeriose, importante doença transmitida por alimentos que acomete humanos e animais podendo causar morte. Foram coletadas 100 amostras de salsicha de mercados do Distrito Federal. A metodologia usada para o isolamento foi a descrita na Instrução Normativa Nº 40 do Ministério da Agricultura, Pecuária e Abastecimento. As amostras que apresentaram motilidade característica de Listeria sp p. foram submetidas a técnica de PCR para confirmação da espécie L. monocytogenes. Os resultados positivos para Listeria sp p. foram vistos em 14 amostras e em 5 confirmou-se a presença de L. monocytogenes. Através dos resultados desse experimento conclui-se que salsichas podem estar contaminadas com bactérias do gênero Listeria sp p. e L. monocytogenes, oferecendo risco de listeriose aos consumidores.

ABSTRACT

O queijo mussarela, tipo mais utilizado no Brasil em preparações culinárias especialmente por características como fatiamento e derretimento, está sujeito a contaminações microbiológicas durante todo o processo que o leva à mesa do consumidor. Diante disso, objetivou-se pesquisar Salmonella spp. e Listeria monocytogenes em queijo mussarela fatiado comercializado em hipermercados de Recife-PE. Foram analisadas quarenta e nove amostras de queijo mussarela fatiado e todas foram negativas para os microrganismos pesquisados. Porém, observou-se Listeria innocuaem 4,1% das amostras (2/49). Estes resultados sugerem provável falha de higienização no local de fatiamento ou embalagem do produto, sendo necessárias ações que garantam a inocuidade dos alimentos ofertados ao consumidor, de forma a não pôr em risco a saúde pública.

ABSTRACT The herein presented assay provided a bacteriological and molecular characterization of 100 samples of L. monocytogenes isolated from human (43) and food (57) sources, from several regions of Brazil, and collected between 1975 and 2013. Antigenic characterization defined 49% of serotype 4b samples, followed by 28% of serotype 1/2b, 14% of serotype 1/2c, 8% of serotype 1/2a, and 1% of serotype 3b. Both type of samples from human and food origin express the same serotype distribution. Multiplex PCR analysis showed 13 strains of type 4b with the amplification profile 4b-VI (Variant I). Virulence genes hly, inlA, inlB, inlC, inlJ, actA, plcA, and prfA were detected in all samples, highlighting a deletion of 105pb on the actA gene in 23% of serotype 4b samples. Macrorestriction profile with ApaI at PFGE showed 55 pulsotypes, with the occurrence of the same pulsotype in hospitalized patients in São Paulo in 1992 and 1997, and two other highly related pulsotypes in patients hospitalized in Rio de Janeiro in 2008. Recognized pulsotypes in listeriosis cases have also been detected in food. Thus, the prevalence of a serotype and the persistence of certain pulsotypes herald future problems.

ABSTRACT This study aimed to investigate the effect of peracetic acid (PAA, 0.5%) on adherent cells of three strains of Listeria monocytogenes strains belonging to serotypes 4b and 1/2b that had been previously isolated from the environment of a Brazilian cheese plant. The assays were conducted using polystyrene microplates and stainless steel coupons and the adhered cells were treated with PAA for 60, 120 and 180 s. On stainless steel, biofilms were partially inactivated by PAA after 60 s and almost 100% of the cells were damaged within 180 s using epifluorescence microscopy with LIVE/DEAD® staining. On polystyrene microplates, PAA decreased (P<0.05) biofilm biomass produced by the three L. monocytogenes isolates at 60 s, when compared with controls (no PAA treatment). However, PAA did not completely eliminate L. monocytogenes cells on polystyrene microplates (decreasing 1.8-2.5 log cycles after treatment with PAA for 180 s). The correct concentration and contact time of PAA is critical for eliminating biofilms formed by L. monocytogenes on stainless steel surfaces, although further studies are needed for defining efficient PAA treatments to remove adherent cells of this pathogen on plastic polymers

ABSTRACT

Listeria monocytogenes é um micro-organismo patógeno que surgiu na década de 80, responsável pela doença de origem alimentar chamada listeriose. A listeriose afeta principalmente mulheres grávidas, recém-nascidos, idosos e pessoas imunodeprimidas. Essa doença pode ocasionar abortos, septicemias, meningites e até mesmo óbito nos casos mais graves. A bactéria Listeria apresenta doses infectantes muito baixas possibilitando que uma simples contaminação ou deficiência de processos que visam à eliminação desse micro-organismo ocasionem surtos. A alta taxa de mortalidade desperta atenção especial das autoridades governamentais responsáveis pelo controle sanitário e da comunidade científica da área de alimentos, mas no Brasil não há estatísticas oficiais de casos de listeriose, pois sua notificação não é obrigatória. Nesta revisão procurou-se trazer à luz informações sobre a listeriose, uma doença de origem alimentar ainda pouco conhecida, identificando os alimentos comumente envolvidos nos surtos, os fatores causais mais significativos, assim como as características e impactos sociais relacionados a essa doença. Foram descritos ainda sua etiologia, fatores de riscos, sinais e sintomas. A análise crítica e a divulgação dos principais aspectos relacionados das doenças transmitidas por alimentos pode ser um importante fator para a prevenção dessas doenças.

ABSTRACT

Listeria monocytogenes is an aerobic, motile, gram positive bacillus recognized as an intercellular pathogen in human where it most frequently affects neonates, pregnant women, elderly patients, and immunosuppressed individuals as well as healthy persons. Ocular listeriosis is rare, most frequently in the form of conjunctivitis, but has been also shown to cause rarely endophthalmitis with pigmented hypopyon and elevated intraocular pressure such as in our case. We are reporting one immunocompetent patient presenting with dark hypopyon following laser refractive procedure. His clinical findings, investigations, and further management are all described with relevant literature review of similar cases. Diagnosis of ocular listeriosis was confirmed by positive culture of anterior chamber [AC] aspirate with identification of the above organism. His visual outcome was satisfactory with good preserved vision. We believe that his ocular infection was exogenous and that ophthalmologists should be aware of the causative organisms of colored hypopyon to avoid delayed diagnosis

ABSTRACT

Listeria monocytogenesis a facultative intracellular pathogen, ubiquitous and aetiological agent of listeriosis. The main way of acquisition is the consumption of contaminated food and can cause serious medical conditions such as septicemia, meningitis and gastroenteritis, especially in children, immunocompromised individuals and seniors and abortions in pregnant women. An increase in cases of listeriosis worldwide has been reported and it is estimated that its prevalence in developed countries is in the range of 2 to 15 cases per one million population. This microorganism is characterized for the transition from the environment into the eukaryotic cell. Several virulence factors have been involved in the intracellular cycle that are regulated, pimarilly, by the PrfA protein, which in turn is regulated by different mechanisms operating at the transcriptional, translational and post-translational levels. Additionally, other regulatory mechanisms have been described as sigma factor, system VirR/S and antisense RNA, but PrfA is the most important control mechanism and is required for the expression of essential virulence factors for the intracellular cycle.


Listeria monocytogeneses un patógeno intracelular facultativo, ubicuo y agente etiológico de listeriosis. La principal vía de adquisición es el consumo de alimentos contaminados, pudiendo ocasionar cuadros clínicos muy graves como septicemia, meningitis y gastroenteritis, especialmente en niños, individuos inmunocomprometidos y de la tercera edad, y aborto en mujeres embarazadas. Se ha informado un aumento en los casos de listeriosis a escala mundial y se estima que su frecuencia en los países desarrollados está en un rango de 2 a 15 casos por millón de habitantes. Este microorganismo se caracteriza por realizar una transición desde el medio ambiente hacia la célula eucariota. Para este proceso se han descrito varios factores de virulencia, los cuales están involucrados en el ciclo intracelular y están regulados, principalmente, por la proteína PrfA, la cual a su vez está regulada por diferentes mecanismos que actúan a nivel transcripcional, traduccional y post-traduccional. Además, se han descrito otros mecanismos regulatorios como: factor Sigma, sistema VirR/S y ARN sin sentido. No obstante, PrfA es el mecanismo de control más importante y el cual es requerido para la expresión de los factores de virulencia esenciales para el ciclo intracelular.

Foram colhidas amostras de 100 carcaças em um frigorífico exportador, localizado no interior do estado de São Paulo, amostradas ao longo de um ano, por meio do método de esponjas, aplicado na região do peito do animal. As amostras foram colhidas em três pontos, denominados A, B e C, sendo cada carcaça amostrada nos três pontos, localizados nas etapas: pós-sangria (A); pós-esfola (B) e pós-lavagem (C). Foram realizadas pesquisas de Listeria sp., E. coli O157, Salmonella spp. e microrganismos indicadores (Petrifilms® AC, EC e EB). Não foram isolados Listeria ou E. coli O157 em nenhuma das 300 amostras. Salmonella spp. foi isolada em nove, sendo oito no ponto A e uma no ponto B. Para mesófilos, as contagens variaram de 0 a 6,8 log UFC/cm², para coliformes totais, de 0 a 4,57 log UFC/cm², e para E. coli, de 0 a 4,38 log UFC/cm². Diante dos resultados obtidos e em comparação com a literatura, conclui-se que o estabelecimento estudado apresenta qualidade, tanto sanitária (devido às baixas prevalências dos patógenos) quanto higiênica (devido à acentuada diminuição da carga microbiana de indicadores ao longo da linha).


Samples were collected from 100 carcasses in a slaughterhouse exporter, located within the State of São Paulo, sampled over a year through the sponge method, applied to the chest of the animal. Samples were taken at three points, denominated A, B and C, each carcass sampled at three points located in the following steps: after bleeding (A) after skinning (B) and after washing (C). Research was conducted for Listeria sp., E. coli O157, Salmonella spp. and Micro-organism (Petrifilms ® AC, EC and EB). Listeria or E. coli O157 were not isolated in any of the 300 samples. Salmonella spp. was isolated in nine, eight at point A and one at point B. For Mesophiles, scores ranged from 0 to 6.8 log UFC/cm²; for Total coliforms, 0 to 4.57 log UFC/cm² and E. coli from 0 to 4.38 log UFC/cm². With the results obtained and compared with the literature, it is concluded that the establishment in this study has both sanitary quality (due to the low prevalence of pathogens) and hygienic quality (due to the sharp decrease in the microbial load of indicators along the line.

Introdução - A bactéria Listeria monocytogenes é um agente zoonótico transmitido, principalmente, por alimentos. Dentre as fontes de contaminação, destacam-se os produtos de origem láctea, carnes e embutidos, além dos ambientes da indústria de processamento alimentício. Na última década, foram detectadas cepas de L. monocytogenes e L. innocua, em ambiente de frigoríficos e alimento. Estas apresentavam variação na intensidade da virulência para células eucarióticas decorrente de mutações nos genes de virulência. Esta alteração em ambas as espécies, e o relato de um caso fatal de listeriose humana ocasionada por L. innocua atípica demandam atenção, pois apresentam maior risco à saúde da população exposta a estes ambientes e alimentos tornando-se, portanto, uma importante questão de saúde pública. Objetivo - Pesquisar genes de virulência em cepas de L. monocytogenes e L. innocua, isoladas em pontos da linha de abate suíno e do comércio de carne no Estado de São Paulo. Material e Métodos Foram estudadas 40 cepas, dentre estas, isolados de L. monocytogenes e L. innocua com atividade hemolítica atípica. Foram realizados testes de atividade hemolítica e produção de fosfolipase A para caracterização dos isolados. A detecção dos genes de virulência foi realizada através da reação em cadeia pela polimerase (PCR). Para confirmação das sequências amplificadas e a análise das mesmas, os fragmentos obtidos foram sequenciados. A identificação molecular das espécies foi realizada por análise filogenética dos genes prs e 16S rRNA. Resultados Dos 40 isolados, cinco de L. monocytogenes e sete de L. innocua apresentaram atividade hemolítica atípica, sendo que nestes últimos também foi observado halo atípico no meio ALOA. As cepas de L. monocytogenes foram positivas para a detecção de todos os genes de virulência estudados. Dois dos isolados atípicos de L. innocua também foram positivos para todos os genes e os outros cinco foram positivos para hly, plcA e inlC. Foram detectadas mutações nas proteínas InlC, InlB, InlA, PI-PLC, PC-PLC e PrfA, nas cepas atípicas, que resultaram em alterações nas suas estruturas secundárias que podem explicar o fenótipo desses isolados. A confirmação de espécie apenas foi alcançada com a análise filogenética do 16S rRNA. Conclusões A partir desses resultados, foi proposta a utilização dos genes prfA, plcB e inlB, como forma de triagem, para diferenciar as espécies L. monocytogenes e L. innocua, de modo a complementar os testes fenotípicos.

ABSTRACT

Majority of studies concerning the gene expression of Listeria monocytogenes have been done on pure culture states. Our objective was to study L.monocytogenes in a co-cultured state and to understand if microbes in their natural state of existence are different in their expression than that of the purely cultured lab grown forms. For a long period discussions have been on the expression of prfA, (which is a virulence gene regulator) in a mammalian host and its role in causing the switch from a saprophytic to pathogenic form of L.monocytogenes. We, in this paper for the first time report the expression of prfA and other virulence genes by L.monocytogenes under different extracellular conditions, and also as a pure culture biofilms, that is different from the previous reports. We also report that the expression of prfA seems to vary considerably when co-cultured with Bacillus subtilis.

ABSTRACT

Invasion of hepatocytes by Listeria monocytogenes (LM) and Salmonella Typhimurium (ST) can stimulate tumor necrosis factor alpha (TNF-α) release and induce apoptosis. In this study, we compared the behavior of hepatocytes invaded by three L. monocytogenes serotypes (LM-4a, LM-4b and LM-1/2a) and by ST to understand which bacterium is more effective in the infectious process. We quantified TNF-α release by ELISA, apoptosis rates by annexin V (early apoptosis) and TUNEL (late apoptosis) techniques. The cell morphology was studied too. TNF-α release rate was highest in ST-invaded hepatocytes. ST and LM-1/2a induced the highest apoptosis production rates evaluated by TUNEL. LM-4b produced the highest apoptosis rate measured by annexin. Invaded hepatocytes presented various morphological alterations. Overall, LM-4b and LM-1/2a proved to be the most efficient at cell invasion, although ST adapted faster to the environment and induced earlier hepatocyte TNF-α release.


A invasão de hepatócitos por Listeria monocytogenes (LM) e Salmonella Typhimurium (ST) pode estimular a liberação do Fator de Necrose Tumoral (TNF-α) e induzir a apoptose celular. Neste estudo comparamos o comportamento de hepatócitos invadidos por três sorotipos de L. monocytogenes (LM-4a, LM-4b e LM-1/2a) e por ST para entender qual bacteria é mais efetiva no processo infeccioso. Nós quantificamos a liberação de TNF-α pelos hepatócitos por ELISA e as taxas de apoptose pelas técnicas de anexina V (apoptose precoce) e TUNEL (apoptose tardia). A morfologia das células foi estudada também. A taxa de liberação de TNF-α foi mais alta em hepatócitos invadidos por ST. ST e LM-1/2a induziram as maiores taxas de apoptose pelo método TUNEL, enquanto LM-4b produziu as maiores taxas de apoptose por anexina V. Os hepatócitos invadidos apresentaram várias alterações morfológicas. Na análise do conjunto de dados, os sorotipos LM-4b e LM-1/2a provaram ser os mais eficientes na invasão celular, enquanto que ST adaptou-se mais rápido ao meio e induziu a liberação precoce de TNF-α pelos hepatócitos.

ABSTRACT

Este trabalho teve como objetivo verificar a ocorrência de Listeria monocytogenes em salames artesanais, elaborados por pequenas indústrias do setor de carnes do sul do Brasil. Das cinqüenta amostras de salames de diferentes fabricantes avaliadas, em 8% foi verificada a presença de L. monocytogenes. Os valores de pH dessas amostras variaram entre 4,35 e 5,99 e a atividade de água de 0,89 a 0,91. A implantação das Boas Práticas de Fabricação neste setor é fundamental para obter alimentos prontos para o consumo e seguros à saúde do consumidor.

ABSTRACT

El principal factor que interviene en el origen y prevención de las enfermedades trasmitidas por los alimentos es la higiene alimentaria. Dichas enfermedades son causadas por la ingestión de alimentos o agua contaminados con microorganismos patógenos ocasionando una infección o por la ingestión de alimentos contaminados con toxinas. Los principales agentes involucrados son Escherichia Coli, Campylobacter, Salmonella, Shigella, Listeria Monocytogenes, Norovirus, virus Hepatitis A, Astrovirus, Rotavirus, y Virus Coxsackie. Toxinas producidas por hongos o por microflora marina y los contaminantes orgánicos persistentes pueden también causar serios problemas de salud. La inocuidad alimentaría ha tomado relevancia debido a una mayor exigencia por consumidores cada día más informados y por las demandas del comercio exterior. Medidas que aseguren una adecuada higiene alimentaría nos permitirá prevenir enfermedades, principalmente digestivas, causadas por variados agentes en los alimentos. Esto se logra por la implementación de las medidas propuestas por la Comisión Internacional conocida como Codex Alimentarius


Food Safety is the main factor involved in the origin and prevention of Food-borne diseases. These diseases are caused by either the intake of contaminated foods or water or by the intake of toxin-contaminated foods. Escherichia Coli, Campylobacter, Salmonella, Shigella and Listeria Monocytogenes, Norovirus, Hepatitis A virus, astrovirus, rotavirus, and coxsackie virus are the main causative agents involved in food-borne diseases. Toxins produced by fungi or marine microflora and the presence of persistent organic polluting agents can also cause serious health problems. Food safety has become an important topic due to a more demanding and informed consumer and the foreign food trade. Measures leading to ensure a better food safety will allow us to prevent various foodborne preventive diseases, mostly in the digestive system, caused by different etiological agents. This can be achieved through the implementation of the different measures proposed by the international commission called Codex Alimentarius

ABSTRACT

Listeria monocytogenes is a foodborne pathogen that causes serious invasive illness, mainly in certain well-defined high-risk groups, including immunocompromised patients, pregnant women and neonates. L. monocytogenes primarily causes abortion, septicaemia or infections of the central nervous systems. Listeriosis outbreaks have mostly been linked to consumption of raw milk or cheese made of unpasteurized milk. Previous outbreaks of listeriosis have been linked to a variety of foods especially processed meats (such as hot dogs, deli meats, and páté). The public health importance of listeriosis is not always recognized, particularly since listeriosis is a relatively rare disease compared with other common foodborne illnesses such as salmonellosis or botulism. However, because of its high case fatality rate, listeriosis ranks among the most frequent causes of death due to foodborne illness: second after salmonellosis. Changes in the manner food is produced, distributed and stored have created the potential for widespread outbreaks involving many countries. The pasteurization of raw milk, which destroys L. monocytogenes, does not eliminate later risk ofL. monocytogenes contamination in dairy producís. Extensive work has been ongoing in many countries during the last decade to prevent outbreaks and decrease the incidence of listeriosis. A marked reduction has occurred in its incidence in some of these countries during the 1990s, suggesting a relationship between preventive measures and reduction on human cases listeriosis.


Listeria monocytogenes es un patógeno que cuando se encuentra en los alimentos, puede causar serias enfermedades, principalmente en grupos de alto riesgo como inmunocomprometidos, mujeres embarazadas y neonatos. Listeria monocytogenes es una importante causa de abortos, septicemia o infección del SNC. Antiguamente, los brotes se vinculaban con una gran variedad de alimentos, especialmente carnes procesadas (salchichas, paté, productos precocidos); en la actualidad, la mayoría se vinculan al consumo de leche cruda o quesos elaborados con leche sin pasteurizar. La importancia de la listeriosis para la salud pública no siempre es reconocida, sobre todo porque es una enfermedad relativamente rara, en comparación con otras más comunes como la salmonelosis. Sin embargo, debido a su alta tasa de letalidad, la listeriosis se encuentra entre las causas más frecuentes de muerte por enfermedades transmitidas por alimentos ocupando la segunda posición, después de la salmonelosis. Los cambios en la forma de producción, almacenamiento y distribución de los alimentos han propiciado diversos brotes. En las últimas décadas, muchos países han encaminado intensos trabajos hacia la prevención de la listeriosis; la notable reducción de su incidencia en la década del '90, sugiere una relación positiva con las medidas implementadas.

ABSTRACT

Listeria monocytogenes, rare pathogen in the general population, causes serious infections in patients at the extreme ages of life, pregnant woman, and those with immunosuppression. The clinical manifestations are essential to suspect the disease in patients at risk, allowing an early prescription of antimicrobial therapy, before the results of the cultures are available. Clinical course and prognosis depends on how early treatment is started and, in pregnant women, the gestational age. In Clínica Alemana, at Santiago, we detected a 15 fold rate rise of neonatal listeriosis between year 2007 and 2008. Ten cases were diagnosed between January and July 2008 and the seven cases occurring in pregnant women are reported here. All these patients were in their first pregnancy, which could be associated with similar lifestyle and food habits. Considering this new epidemiological scenario, it is important to educate the population, and to conduct an epidemiological study in order to determine the national situation of Listeria monocytogenes infection.


Listeria monocytogenes, es un patógeno poco frecuente en la población general, causante de infecciones graves en pacientes en edades extremas de la vida, mujeres embarazadas e inmunodeprimidos. La sospecha de la enfermedad en pacientes de riesgo se basa principalmente en el cuadro clínico, lo que permite iniciar un tratamiento empírico antes de contar con los resultados de los cultivos. La evolución y pronóstico dependen de la precocidad con que se inicia la terapia y de la edad gestacional. En Clínica Alemana de Santiago detectamos un aumento de 15 veces en la tasa de listeriosis comparando el año 2007 con el 2008. Entre enero y julio 2008, se diagnosticaron 10 casos, de los cuales siete fueron en primigestas, lo que podría tener relación con un hábito alimentario y características de vida similar. Es fundamental, a la vista de esta nueva realidad epidemiológica, educar a la población en hábitos alimentarios y de higiene, como también realizar un estudio epidemiológico que determine la situación nacional de infección por L. monocytogenes.
